Mass Effect and Fallout are western RPG's that go into the shooter genre, both let you make choices in the quests/missions you carry out, making the good or evil choice
the difference is that Fallout is more of a sandbox game like the GTA series and Mass Effect is pretty much go from point A to point B (or C, D, E, F if you wish)
the story in both are pretty good although Mass Effort is going to be a trilogy with the second game coming out early 2010, if you want Fallout has some good DLC especially Broken Steel since it lets you continue playing after the main story is done and up the level cap from 20 to 30
if i had to pick between the two I would take Fallout 3 but thats if you want to get into the DLC's, if not, go with Mass Effect since you can get it for 20 brand new
i did forgot that the fallout 3 game of the year edition does come with all the DLC's that been released so far
